Hello,
For the moment we are running Advanced Analysis V1.4SP6 and we would like to upgrade
to V2.1SP3.
Is it necessary to uninstall first the V1.4 or not ? And this for client and server add-on ?

It's not the worst idea to do an uninstall before installing new AO versions. We had some problems in past that version number changed but not all features are working without the uninstall.
No, these are just updates; no need to uninstall unless you experience issues
