In one of my Custom form i have a matrix. when i click matrix header, the column is selected. how can i deselect the column ?

You can not deselect a column. The only thing you can do is to select another column (which will "deselect" the prevouis column)
You may deselect a column by either clicking that column while holding the Ctrl key, or by clicking on the top left square, which will deselect everything.
